Collect one green top (sodium or lithium heparin) tube and one purple top (EDTA tube).
Collect 10 mL of whole blood and two (2) foam-tipped buccal swabs prior to food or drink, or after thoroughly rinsing mouth with water.
Additionally, please send one normal control (heparin green top tube) from a healthy, unrelated individual.

Do not centrifuge! Keep at room temp (18-22°C).

Frozen, clotted, hemolyzed or centrifuged specimens. Anticoagulants other than heparin. Specimens >24 hrs old.
Maintain and transport specimens at room temperature (18-22°C).
Ship specimens Priority Overnight via Fed Ex in container sufficiently insulated to avoid temperature extremes (<10°C and >37°C).

Mutations of the gene encoding STAT3 are associated with the Hyper IgE syndrome (HIES). Because development of TH17 cells is dependent on STAT3, patients with HIES may have depressed levels of TH17 cells, thereby resulting in a compromised ability to fight infections. Analysis of the percentage of circulating TH17 cells assists in the evaluation of HIES.
