Description
This test code is for identification of a positive AFB culture.
The laboratory should be notified when the presence of M. genavense, M. haemophilum, or any other fastidious mycobacteria are suspected, as these organisms will not grow on media routinely used for subculture.
If colony morphology on a solid medium resembles Mycobacterium tuberculosis (MTB) complex, a line probe assay to detect M. tuberculosis complex will be performed. If positive, first-line drug antimicrobial susceptibility testing will be performed on all initial isolates. The laboratory will provide a subculture to the respective state laboratory for genotyping upon request (at no charge).
All other mycobacteria will be identified using line probe assay or DNA gene sequencing (16S or rpoB genes).
Identification performed by a combination of the following methods as determined by the National Jewish Health Infectious Disease Mycobacteriology Laboratory:
Line Probe Assay Identification of Nontuberculous mycobacteria including M. avium, M. chimaera, M. chelonae, M. abscessus ssp abscessus, M. abscessus ssp bolletii, M. abscessus ssp massiliense.
rpoB gene sequencing for identification of mycobacteria.
16S gene sequencing for identification of mycobacteria and aerobic actinomycetes.
Line Probe Assay Indentification of MTB complex species including M. tuberculosis, M. africanum, M. bovis ssp bovis, and bacilli Calmette-Guerin (BCG).
