Rheumatoid arthritis (RA) is a systemic condition affecting approximately 1% of the general population leading to progressive arthritis and extra-articular manifestations (ExRA), including interstitial lung disease (RA-ILD). The RA-ILD manifestations become more prevalent as RA progresses despite the use of therapies, which effectively treat the arthritis. We have developed a new mouse model to define the mechanisms leading to RA-ILD and discover unique biomarkers, which could predict the development and lead to earlier treatment of patients with RA-ILD.

We also are studying how pregnancy impacts maternal antigen-presenting cells (APCs) and therefore T-cell function to establish new insights into the mechanism by which pregnancy suppresses RA. The knowledge gained from these studies should increase our capacity to manipulate the immune system to treat both the articular and extra-articular manifestations of RA. We are accomplishing the goals of these studies through basic investigation using novel mouse models we generated as well as translational studies using samples from subjects with RA, RA-ILD, and controls.
 
We also are pursuing new strategies to manipulate lymphocyte-signaling pathways to treat autoimmune disease. We are defining how alterations in ubiquitinylation as well as neddylation alter signaling through the T-cell receptor (TCR) complex in mouse models of inflammatory arthritis to determine their role in autoimmune pathogenesis. We have shown that the loss of an adaptor of the E3 ubiquitin ligase c-Cbl leads to the enhanced develop and function of Tregs, which suppress the development of arthritis in the SKG mouse model. We has also shown that a neddylation inhibitor decreases the threshold for TCR signaling leading to enhanced IL-2 production in T cells and the arrest of progression of arthritis in SKG mice.  We are actively defining the mechanisms that govern these observations with the goal of our studies providing foundational support for developing drugs to target ubiquitination and/or neddylation to treat autoimmune disease.
